چکیده

Neutral fucolipids and fucogangliosides of normal rat liver, normal rat hepatocytes, and rat hepatoma H35 and HTC cells have been compared. H35 cells were characterized by accumulation of a relatively large quantity of a few neutral fucolipids and a major fucoganglioside. HTC cells were characterized by accumulation of several neutral fucolipids and by the absence of fucogangliosides. Those fucolipids which accumulated in hepatoma cells were absent in normal rat liver and normal hepatocytes. Normal hepatocytes and normal rat liver contained a low quantity of fucolipids which migrated differently on thin-layer chromatography from those which accumulated in hepatoma cells. In addition to these qualitative differences, the quantity of total fucose in purified glycolipid fraction was much higher in H35 and significantly higher in HTC cells than that in rat liver. A fucoganglioside which accumulated in H35 cells was tentatively characterized as a fucosylated GM1 ganglioside with the following structure: (formula: see text). The results indicate that anomalous fucosylation on glycolipid may take place in hepatoma cells.

Effect of insulin, dexamethasone, and glucagon on the amino acid transport ability of four rat hepatoma cell lines and rat hepatocytes in culture.

The basal level and hormone-affectedaminoacidtrans portof rat hepatocytesand rat hepatomacell lines H4-bI E-C3 (H35), HTC, McA-RH8994 (8994), and McA-RH7777 (7777) in culture were examined by investigating the amount of a-[1-14C]-aminoisobutyricacid (AIB) trans ported in 4 mm and up to 4 hr at 37°. The substances testedwere insulin, dexamethasone(DEX),glucagon, and dibutyryl cyclic adenosine 3' ...

HEPATOMA McARDLE-RH7777 CELLS HAVE THE SAME RESPONSE AS NORMAL RAT HEPATOCYTES TO BOTH DIBUTYRYL-cAMP AND ANTICALMODULINW-7

The effects of cAMP-analogue dibutyryl-cAMP and anticalmodulin W-7 were studied on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle RH7777 cells and normal rat hepatocytes. Dibutyryl-cAMP and W -7 separately caused a significant decrease in the secretion of de novo synthesized triacyl [3H]glycerol in both cultures of McArdle cells and rat hepatocytes. The inhibito...

Comparative studies of glucose metabolism in HTC, RLC, MH1C1, and Reuber H35 rat hepatoma cells.

Carbohydrate metabolism by four rat hepatoma cell lines in culture, namely, Reuber H35, MH1C1, RLC, and HTC, has been investigated. Glucose utilization by H35 and MH1C1 cells is lower than that by RLC and HTC cells. The four cell lines also differ with respect to the accumulation of lactic acid in the growth medium; in particular, H35 cells show uptake of lactic acid, rather than accumulation i...

Expression of CXC Chemokines Gro/KC and SDF-1a in Rat H4 Hepatoma Cells in Response to Different Stimuli

Background: It is now well established that several environmental stress factors cause activation of p38 MAP kinase and JNK in various cell types to produce chemokines. Objective: To investigate the expression of CXC chemokines Gro/KC and SDF- 1a in rat's H4 hepatoma cells in response to heat shock, hyperosmolarity and oxidative stress. Methods: Hepatoma cells were maintained in MEM medium. Cel...

Regulation of Phosphoenolpyruvate Carboxykinase and Tyrosine Transaminase in Hepatoma Cell Cultures

The ability of N(6), O(2)'-dibutyryl cyclic AMP (DBcAMP) to regulate a number of metabolic events in four lines of cultured rat hepatomas has been examined. Although dexamethasone induces tyrosine transaminase in all four lines, DBcAMP induces this enzyme normally only in H35 cells. A slight increase in transaminase activity was seen with MH(1)C(1) cells and HTC cells, but no effect was detecta...
